The computer that runs the UEM Management Console needs to be able to browse the domain. So it should be domain joined and a domain user should be logged on.
If you have multiple domains, make sure the trusts are correctly placed.
Maybe re-joining the computer to the domain can solve the problem.
Do you also have this problem if you run the UEM Management Console on another computer?

This problem occurs in other VM also. i have manually set AD entry in hosts file for ADC created in same VLAN and Same Datacenter, still this issue is happening. if i run ipconfig /flushdns and search OU with entry vmware.com it works but later same issue happen after few hours.

This seems like a DNS problem or some other AD problem.
UEM just leverages the Windows capabilities and does nothing special.
