First upgrade vCenter Server and all his plugin.

Then you can use VUM to update the ESX.

If you do not have vCenter Server, you can use the upgrade zip file and the esxupdate utility to patch each ESX.

I have already upgrade my VC to Up[date 1. When i try to upgrade my ESX host using Update manager i receive the following error. "Upgrade not supported for Host" Did you use the Zip file or the iso file to upgrade your host?

You do not need the zip file with VUM (it can be used with esxupdate).

The U1 is only a set of patch of the vanilla 4.0.

So use the same baseline that you have use with 4.0 and you will see that your ESX 4.0 become a U1 version.
