Can i Scan the host while Host in Running

I have 5 hosts in my environment i need to scan the host in Update manager do i need to put the host in Maintenance mode  or not.

Nope.  You can scan the hosts at anytime and not impact system availability.  If you want to patch them, you can put them in maintenance mode, but Update Manager can automate that part of the deployment of patches as well.
