i use standalone ESXi 5.1U3 in my network that contain 10 VMs
and it's run on HP DL380 G8.
I'm planning to upgrade and download HP Custom Image ISO for ESXi 6.0U2.
nowadays HP release "HP Service Pack for ProLiant (SPP) 2016.04.0".
i'm confused, is it neccesarry to use HP SPP or not?
if Yes, before upgrade or after.
or just upgrade with only HP Custom Image ISO?

The HP SPP is basically just a huge firmware and HP software update live boot DVD. It does not contain ESXi binaries. You can boot your server from the SPP to automatically update the firmware components, but you can also do it online in ESXi by downloading individual firmware update packages that can be executed from the ESXi shell.
It doesn't really matter but I would update the firmware beforehand, make sure everything works right, and then update the actual ESXi (preferably with the HP ESXi image).
HP OEM Custom VMware Image contains particular version of ESXI and driveres related to HP Hardware. This one will never upgrade firmware
HP SPP contains fimware and driver upgrades for HP hardware.
Correct procedure will be:
* Upgrade ESXi 6 from HP Custom Image.
* Upgrade Firmware (either offline by booting to the SPP or online from within ESXi)
* Upgrade Driveres (Online from within ESXi)

can you tell me what command updates driver for ESXi?
You can either manually download driver vibs to your ESXi host and than install by "esxcli software vib update _package_" (not sure where exactly are the vibs stored on the CD).
Or
Launch HP SUM from the SPP and scan your inventory (ESXi host) to automatically deploy the drivers and firmwares
