We currently have VSphere 5.5 Enteprise Plus and planning on upgrading directly to VSphere 6 with Operations Management. Is this a one time upgrade path or is it upgrade to VSphere 6 and then install Operations Manager after?

The vSphere with Operation Management is a "suite" that include the vSphere ESXi (Standard, Enterprise or Enterprise Plus) and the vRealize Operations Standard (maybe with some additional)... this way, you can treat the vSphere ESXi upgrade in the same way... and about the vRealize Operations, you can install later, that some features are distributed on virtual appliance format.
