Google is your friend:

vSphere Virtual Machine Replication: VMware | VMware United Kingdom

vSphere Data Protection & Recovery: VMware | VMware United Kingdom

The first is a help for DR, the second is for backup/restore.

Do vSphere Replication and Data Protection perform their jobs on the currently running live VMs or on the VM datastore file ?

From my current understanding, vSphere Replication would be performed on the live VMs, while Data Protection performed on the VM datastore file. Is this correct ?

The answers to those questions (and many more) can be found on the pages I linked to.

Both vSR and vDP take data from the datastore of the VMs.
