Yes, ESXi fully supports SAN and NAS. Setup process is very easy and described in details in manual.
So, actually you're planning to have 3 hosts with local storage, not with SAN or NAS (shared storage)?

Yes Planning to have 3 ESXi 4.0 hosts and shared storage either SAN/NAS. Currently I dont have vCenter installed so all my hosts will be independent.

Depending what kind of software you will run in VM you will have to choose between SAN and NAS. In anyway, I would you recommend to try StarWind iSCSI SAN virtualization tool.

As I know Datacore is pretty complicated, Lefhand is perfect , except the price. Openfiler is free but, commercial aplications are better when stuff is about support.
