You only need to enable Jumbo Frames on your physical switches if you plan to actually use them on your connected end systems, that is your ESXi hosts or in VMs. Unless you manually change the MTU on ESXi vSwitches, vmkernel ports or VM vNICs, no vSphere component (any other OS too) will ever try to communicate via Jumbo Frames.
Typically, you only use Jumbo Frames on an isolated iSCSI network or maybe for the vMotion network but that's about it. The performance benefits of Jumbo Frames for 1G networks are not significant, but on 10G it can provide improvements.
