Storage Vmotion on 3.5u2

After upgrading the vCenter server to v4.0 can I then use storage vmotion on 3.5u2 hosts?

sVMotion should work, but you'll need to ensure you have the proper licensing. sVMotion relies on having the vMotion license and with vSphere you'll need either the Enterprise or Enterprise Plus licensing to have both vMotion and sVMotion functionality.

sVMotion works if you have a license for it. What doesn't work on 3.5 hosts is to separate the vDisks with sVMotion. Only the whole guest can be moved. With an ESX 4 host you can move vDisks independently.
