When I select the ISO file for use by the Host Update Utility, an error appears during validation:

In following the VMware ESX 3.5 to vSphere 4.0 documents, I have:
Updated VMware vCenter software to 4.0.
Updated VMware Infrastructure Client 2.5 to VMware vSphere Client 4.0.
Confirmed that VMware Host Update Utility 4.0 is installed.
Confirmed that .NET 2.0, 3.0, and 3.5 are installed on the computer housing the Host Update Utility.
Restarted that computer following the update.
Downloaded host update media (VMware ESX 4 Update 2 .ISO) and verified the MD5 checksum.
Am I using the correct ISO file? My goal is to update a host from ESX 3.5 to vSphere 4.0.

I think the version of the ISO just needs to be the latest build of ESX 4.0. However are you SURE ESX 3.5 is NOT ESXi? also you can't upgrade ESX to ESXi or vice versa.
Yes, I am sure that I am using ESX, not ESXi.
Are you saying that I need to update ESX 3.5 to ESX 4.0 and then ESX 4.0 to vSphere 4.0?
I tried uninstalling and reinstalling all .NET versions, to no avail.
