I'm planning to move foward with a migration of 3.5 to VSphere, I have 2 questions before.
1. Where is the best place to put the COS vmdks, I have local disk and SAN. Which is better and why?
2. I'm currently using VCB to backup VMs to tape, can I use VMWare Data Recovery to backup to disk, and than backup to tape? Or should I continue to backup with VCB?

Put on local storage.
Otherwise you must create some LUNs for each ESXs cause a COS on a shared storage is not recomended.

You can use VDR to backup to a share and then backup the share content to a tape.
But if you need a backup to tape I suggest to still use VCB.

Also, just do a little reading around the communities before you put all your eggs in the VDR basket. Check out thislong post in particular. I would stick with VCB or move to 3rd party product for backup.
