What's the best way to move my Replication Appliance to a new Datastore?
Can I shut it down and then Storage vMotion or is there another, better, way?

You can move appliance to another storage but it is better way to power off the appliance of stop all services of appliances and must check and remove all necessary things which can stop to storage vMotion of appliance.
