VMware ESXi, 5.5.0, 1331820 to ESXi-5.5.0-20170904001-standard (Build 6480324) patch

hi

I was trying to update  VMware ESXi, 5.5.0, 1331820 to  ESXi-5.5.0-20170904001-standard (Build 6480324), there was lot of updates in between, I have reach to  3343343 and it was working fine. but when I update 2016080001, while doing the sixth update we lost the Vcenter connectivity,   please suggest me the best procedure, is it required to update vcenter first?

Yes, for some of the updates, it's required to update vCenter first, see VMware Product Interoperability Matrices

Btw. how do you patch the hosts? In case you do this from the command line using esxcli, there's no need to install the updates one after the other. Assuming that you have supported hardware, you can directly install the latest patch bundle. Note that there might be restrictions regarding the upgrade, if you are using a vendor/OEM ESXi installation.

Yes, for some of the updates, it's required to update vCenter first, see VMware Product Interoperability Matrices

Btw. how do you patch the hosts? In case you do this from the command line using esxcli, there's no need to install the updates one after the other. Yes, since you have reached ESXi 5.5 Express Patch 9 (3343343) which was realsed ESXi 5.5 Update 3b. You have to upgrade vCenter before upgrading ESXI to Update 3b. As Andre mentioned, check VMware Product Interoperability Matrices
