Very Strange Issue:
We have a Windows 2008 server running vSphere 4.0 at our home office named "vCenter". It manages two vSphere 4.0 esxi boxes called esx1 and esx2 at our colo over a 50 meg point to point. Everything works fine but then over night, we lose connectivity to one of the esxi boxes. The really wierd thing is this....from "vCenter", I can ping the esx1 and esx2 intitially, but when one of the two esx boxes drops, I can no longer ping it from "vCenter". That said, I can still ping esx1 and esx2 from another server,(server A in diagram) on the same segment as vCenter so I know that it is not a routing issue. As soon as I restart the esx box, the ping starts working again. Let it run for a few hours and again, we lose connectivity. Funny Business and I am just lost at this point. Any help would be Greatly Appreciated.

This look like a network issue.
Have you checked any Firewall between vCenter en ESX hosts?
You also have to check out any DNS issue. vCenter Server trusts on DNS service, so you have to check this too.

We ran into a similar issue recently and it turned out to be firewall changes that impacted vCenter server subnet, so I would agree that is a good place to check.
