I had 3 hosts, each host can log in via the vSphere client but can't log in vCenter only.
Already I used over 10 years, and suddenly can't log in vCenter.
Can log in vCenter via Web console and Putty, only can not vSphere client.
I use vCenter, ESXi, and vSphere all same 5.1.0 version.
You should check the viclient log.
But did you try to increase default time out value of the vSphere client?

And one more thing, are connecting to the VC with root password?! why don't you use the admin account?
You likely have an expired certificate. Let me tell you this..... 5.1 was the ABSOLUTE WORST version of ESXi and vCenter when it came to SSl certs. The process was very difficult and if something went wrong you were stuck. They have made significant changes since then.
As such, I am not going to provide steps to look and repair certs on this version. You really should work with support on this one, or get off 5.1. Why are you running such an old version that has no more security updates available?
I use this server for internal networks only.

I wish I could help. 5.1 is very old and none of the tools I want you to run will work.
VDT is not supported / lsdoctor is not suppoted either.
I really think you need to open a case with support (pay if you have to). 5.1 had a very difficult process to replace the certificates. Also 5.1 doesn't even have a webclient, you are still using the old C+ client to access vCenter.
I notice you are logging in as root, can you try and login as administrator@vsphere.local (hopefully you have the SSO password)
I am going to assume this is running on Windows ? I don't think they had the appliance in 5.1 , if they did it was one of the first iterations.
