We have customer having two site.
On main site they have one vcenter & two ESXI host (Cluster) and on DR site they have vcenter & standalone host.
Can we configure the High availability between two different sites so if two host in main site is down then VM will failover to DR site host.
Please let us if it is possible to configure the HA between two sites in VMware 6.5

For vCenter HA, We need to Create a Separate port group for vCenter HA network. This vCenter HA network is used for internal communication between the vCenter HA nodes. I hope this internal communication IP of vCenter Should be in the same subnet. Not sure We can create it across different sites.

Yes between sites we can do it through stretched cluster.

Streched cluster works only when you have single storage (streched VSAN or DellEMC VMAX or HDS GAD or whatever else).
In your scenario I would recommend to check vsphere replication or any similar software, Veeam Backup&Replication for example.
But this won't be automatic failover. In case of disaster scenario (whole site down) you will have to push the button to recover VMs.
