does ESXi support Datastores via SMBv3 ? And what would be the prerequisites ?

For a datastore where you can run VMs you need either a block device (via FC, iSCSI or local) or a network file system like NFS.
ESXi can mount CIFS shares, but it will not show up in the datastore overview and you can't use it as a VM location.
