trunking question

in a situation like the one in the image.

do the ports where those physical (vmnick1 and 2) cards connect in the switch have to be in trunk mode ?

Basically yes, assuming you are talking about Cisco trunks (802.1Q), i.e. no channels or LACP. Since the VLAN tagging is done on the virtual port groups, the physical switch ports need to be setup in a way to accept the already tagged traffic, and also forward all traffic to the ESXi host without removing the VLAN tag.

For incoming traffic, the ESXi host will forward the network frames only to those port groups with a VLAN ID that matches the VLAN tag in the frame. The port group is then responsible for removing the VLAN tag an forward the traffic to the VM(s).
