Our customer has a VM which has 10 pieces VMDK. There are some 1Tb size, and there is a large size: 6Tb. Can I create VM snapshot for these large VMDK file? How long can be the snapshot creation time? The whole VM VMDK summary: 12 Tb.

Yes, I want to use the snapshot for backup (Veeam).
go ahead. I have VMs even with greater capacity in the backup. Only Commvault instead of Veeam

I have no problems at all. First backup could take a while but all the rest, I mean in the next days, are working fine.
