I'm trying to troubleshoot one of my mailservers by setting up the config file to run just tile0 and tile4. It begins setting things up and then bombs out with this error message:
Is there any way around this? I don't want to run tiles 1-3 because of overload concerns, just 0 and 4.
I've run the staf command
staf client5 process start command "c:\\Progra~1\\Exchan~1\\LoadGenCmd.exe /r /c C:\\vclient5\\mailserver\\vmmark2-mailserver-5.xml" returnstdout stderrtostdout wait
from the prime client and observed mailserver4 firing up activity. I want to see if I can get good information from it for the analysis. When I run all of the tiles I'm seeing 0s for mailserver4.

Does client4 have a c:\vclient1 directory?
By default you'd have a c:\vclient4 directory. To use out of order clients you'll need to copy it and rename the copy to vclient1.

To expand on what Josh said, running only tiles 0 and 4 would be similar to testing a single failing client-workload pair, as described on page 159 of the VMmark Benchmarking Guide, except that all workloads are running, and the C:\vclient directory number for the out of order client matches its relative tile number as a general rule.
Do I also need to change the reference, in the prime client's hosts file, to this new mailserver1?

It's not necessary for you to change the hosts file on the prime client. Only the VMmark2.config file and the name of the C:\vclient directory must be changed. When you are running an out of order tile, all IP addresses and hostnames on the out of order tile should remain unchanged.
For you, the relevant settings in VMmark2.config would look something like the following. This allows the harness to recognize tile4 as if it were tile1.
