If our Connection cert was to be left to expire, what would the users experience be? Would they not be able to connect to their VDI machines?

It depends on the configuration of the infrastructure and above all on the settings of the Horizon Client or ThinClient
For example on the Horizon CLient you can set how the client should behave in front of an untrusted certificate.

On thinclient there are the same option (for example on teradici thinclient) 

I suggest you replace the certificate, it is a very simple process.
