Hello,
Just let you know that my upgrade is just finished. I upgraded my entire farm with five clusters, 30 ESX Hosts and 100 VM. From ESX 3.01 to ESX3.5U4.
This upgrade taked me about one week. The only thing that I need to do is to install VMware Virtual Tools in the Virtual Machines. Right now, I have 3 approachs:
1-Get in each virtual Machines and Install the VMtools,
2-Install by a GPO from the Domain to the VM,
3-Install from the VCM with the vmware.exe and set the parametres.
I'm waiting for your feedback about these approachs.

I would go with the GPO route. After creating the GPO you should do a couple of VM reboots and ensure the tools updated.
I prefer to use powershell or RVTools. RVTools also gives a nice overview of the current version and status and has a nice gui.
