Is it possible to increase the size of a VMFS partition without using extents in ESX 3 U4? All I can see in the server config manual is to use extents.

No, 3.5 can't extend VMFS partition. The only way - create new datastore of desired size, migrate all VMs and delete old.

Hello. The "VMFS Volume Grow" functionality was added with vSphere. With ESX3 U4, you will have to use extents, which is generally not recommended.
Good Luck!
Not without reformatting. I have done this a few times. Storage vmotion all files. Increase and reformat.
ESX introduces the functionality to increase a VMFS partition.
