ESX3.5 support layer 3 network

Is the ESX3.5 support layer 3 network? I just know that VMotion must be in layer 2, how about another, like console and VM

The vSwitches don't have any awareness of Layer 3 protocols (with limited exceptions).

If my physical network using layer 3 network, and it connect to ESX server, does it work?

Yes it does - if you think about anything that gets an IP address works at Layer 3 of the OSI model - so yes ESX will work with your Layer 3 network -
