Is an upgrade from ESX 3.5 U1 directly to ESX 3.5 U5 supported? Or do I have to go up 1 level at a time?
Thanks in advance
Yes, please refer to the release notes of ESX 3.5u5: http://www.vmware.com/support/vi3/doc/vi3_esx35u5_rel_notes.html
Specifically Upgrading or Migrating to ESX Server 3.5 Update 5 which gives you a nice chart

I am at version 3.5.0 77234 is this U1 can i still upgrade with out damaging VM?
If you take a look at the chart, even base ESX 3.5 is supported. One way to find out is to run:
esxupdate -l query
If you have any Updates, it'll list it out if you're on u1,u2,etc. else you're probably on base 3.5. Again, all 3.5 releases are supported, so you should be fine
