I need to create a 275 GB Drive on a Windows Server 2003 VM. When I select to Add->Hard Disk->Next->Create a new Virtual Disk, it then asks for the size. It will only allow me to set it to 256 GB, not 275 GB.
I know that this is because the default block size is 1MB, but how to I set the block size to 2MB?
You will need to destroy and recreate the datastore to get the choice to change the block size during the creation process.
Move the VMs off first.

I didn't make clear that the new Drive is on a completely NEW LUN. This
is virtual disk #4 for this VM.
The LUN is 350 GB, but I want to create a 275 GB drive on that LUN.

Then you need to wipe the LUN, and recreate the VMFS on it specifying a 2MB blocksize.

The only way I know for you to be able to change the block size is to recreate the vmfs volume.
Just like when you added it the host, Go into the Configuration tab, Select the volume, Remove it.
When you Add Storage to add it back you will be given the choice to set the block size, the 2 meg block size will give you a 512 GB file / disk size limit.
The same applies to a block size of 3 and 4 mb for 1 and 2 TB file / disk size.
Move everything off the volume you are going to remove from the host.
