Question :

Is it possible to Create a connection Server without any direct communication with desktops in order to use multiple domain systems.

Horizon requires a two-way or at a minimum a one-way trust with any domains that you want to authenticate with. The Horizon Agents and Connection Servers also need to communicate on the agent JMS ports.
