What's the best practice for a "regular" VMware server environment and VDI? Can one environment (ESXi and SAN) be used to host both if this is a brand new configuration? Or best to keep separate?

The Quick and dirty answer is that "it depends"
serioulsy, it really depends on two things budget and IO. if you had the money for two SANs then buy two and have one to host your Server Environment and the other to host the VDI desktops, their IO profiles are completely different.
if not, try to keep each use type to their own dedicated LUNs.

As Tom said, it really does depend. How many servers and desktops are you virtualizing? Have you logged any statistics within your server and desktop environment to indentify , CPU, Memory, Disk IO and Network needs - that should give you a decent start. Who is your server and SAN vendor? Do they have any blue print architectures you can review?
