2 Quad Ports per host
Is it possible to have 3-4 ports on Quad card #1 connected to pSwitch A and 3-4 ports Quad card #2 ( in same host) connected to pSwitch B?
Is this required or would the pSwitches handle re-routing?

It is supported - and recommended - to have multiple pNICs per vSwitch with the pNICs connecting to different pSwitches. The vSwitch will use the "Link Status Indicator" on the pNIC to validate connectivity. If connectivity is broken (the link status goes to Off), the vSwitch will automatically re-route the outbound traffic. You can gain additional path failure detection by using beaconing on your vSwitches...
