we are going to start backup of the vmdk files using API and netbackup. So, what all I have to do from vmware side, is that API i have to install or it will be with my setup already. I have ESX 4.0 and Vcenter 4.0 in my environment. Thanks

You need the VMware "agent" on Netbackup side (it isn't a real agent, but only a license option).
Check on Symantec docs on how to configure it.
On VMware side you only need VMs with virtual hardware 7 (to use incremental backup).

okie,so my question is where this vstorage will come into account,that agent which you said will have API for vmware-netbackup integration?and thanks for confirming that virtual machine hardware 7 will server the purpose from vm's and vc side.
API are just embedded on ESX/ESXi and of course vCenter Server (all must be 4.x).
So which a valid account (you can use the default VCB role on vCenter) you can make backup.
On Netbackup you have to configure this creadential and jobs.
There is a good guide on Symantec site, but I cannot find the direct link.

You do not need to install anything on vSphere. Symantec Netbackup will use the CBT (Changed block tracking) feature of vSphere to do block level incremental backups. This just needs to be enabled per VM (unlike other backup solutions Netbackup won't do this on its own).
