Yes, you may use direct upgrade path for 4.1 to 5.1, with update manager or cd installer using of the binary "
VMware-VMvisor-Installer-5.1.0.update01-1065491.x86_64.iso"
This is supported path, you may refer to this release notes. http://www.vmware.com/support/vsphere5/doc/vsphere-esxi-51u1-release-notes.html

While it can be done, it does not always work. In my situation, using Update Manager does not allow me to update directly from 4.1 to 5.x. Instead, I have to boot to the 5.x ISO and do a force migrate. It is something to do with old drivers that no matter what I do, the Update Manager will not cope with properly.
Checking/not checking this box gave me the same result every time; no dice.
The odd part is the VIBs appear to be VMware's own -
This is cause of you adding additional/extra VIB for the current ISO, that are reason which you not be able to used the ISO, but in that case, you may inject the drivers and just ignore the removal. It's will be worked or else, you may find your 5.1 update 1 with oem drivers from the OEM website such as HP,IBM, and etc.
This will be able import into Update Manager and applied the updates.
