I was moving all vm's off one ESX 4.1 server so I could apply updates. All VM's move. On one VM it would not start after it moved with no errors. It said The system cannot find the file specified. The file was the vmdk file. I went to the datastore and the file is there. I tried to copy the file and it said "Unable to access file".
What can I do to get this machine back up again?

In this case I'd recommend you open a call with VMware support to avoid the risk of possible data loss.

Copying the file will not help you to power on the VM because when you add the Hard disk it create the unique id in background. In this case, remove disk from VM(do not select remove from virtual machine and delete disk) and re-add it, power on the machine it will work.
