Is it possibile to use a standard Essential plus key on a DELL customized image?
Is it possible to eventually downgrade from Vsphere 6 to 5?

Yes vSphere licenses can be applied to any image of ESXi, the Dell, HP, Lenovo etc. customized images just include that particular vendor's drivers in the image to save you having to add them to the image yourself or prior to install, it has nothing to do with how the product is licensed.
As for downgrading, you can downgrade your license keys through MyVMware, find your license keys you wish to downgrade and you'll have the option there. To downgrade the software itself if you've already installed v6 then this guide may help you- vCenter doesn't necessarily have to be downgraded as you can manage your v5.x hosts with 6, unless you have to due to licensing constraints.

if it is a valid license , you can activate that on the vsphere editions , try to attach and assign the license
and the question for downgrading the vsphere.. it involves planning and the type of the of environment on which the vsphere is involved ,
1. take a back up of your vcenter data base
2. use the recovery option form DCUI shift+R
3, make sure the bootbank has the desired verison of Hypervisor(if you have choosen the upgarde option ,you will see the previous verison of esxi)
4. you can downgrade the vm hardware version using vmware converter
