So here is one of many questions that will come from me.
I have some lefty users who are used to customizing their mouse to their needs. They need to redo them each time they log out of a machine. Can these settings be saved for them with UEM??

Most certainly! On the Personalization ribbon tab, click Create Config File, pick Use a Windows Common Setting in the wizard, and click Next.
In step 2 of the wizard, select the Mouse option:
Click Next again, pick a suitable file name (Mouse, for instance :-), and click Finish. Voilà!

That is it? Do I need to apply like Directflex or will just creating this setting make it possible??
It's just as simple as that
DirectFlex can only be enabled for executables/applications. Windows settings like Mouse, Wallpaper, etc. don't need DirectFlex.
