As per the DEM logs i can see that folder redirection policy takes like 10 sec to apply it which makes the logon delay. our environment average logon time is 16-20Sec, not more than that. Now the logon takes 33-40 Sec, most of the time wasted in folder redirection., anti virus exclusion already in place. What could be the issue

Do you have condition set or multiple conditions defined for folder redirection? Like a nested group or large condition set?
There no condition. We are using standalone windows file server where profile get redirected.
is it non persistent desktop gets refreshed everytime?
Yes. You are correct. I have case with VMware but no use. They are not able to find the root cause of issue.

You can use the latest version of os optimisation tool + please try disabling Active Setup config file on dem and try logging in
