i download app volumes iso from vmware site. here is in-guest-vhd file in iso file . i want to instal and config app volumes . now i find lots of documents for app volomes but i cant see about this in-guest-vhd. for what ? how to use it and want to publish aplication ( winrar , office etc..) must i this template or they are added feature
These are the template files for your appstacks and writable volumes. If you install the appvolumes manager it will ask you on what VCenter you are working on. It also asks which datastore will be used for Appvolumes (best bet is to go for a SAN here) and after installation it will copy these templates onto this datastore in the folders cloudvolumes\writable_Template and cloudvolumes\appstack_templates.
If you then create an appstack or writable volume what it does is copy one of these 4 files into the cloudvolumes\writable or cloudvolumes\appstack folder where you could actually use it.
But again (as said in another post). I would strongly suggest reading the user guide and try to get a grip of what Appvolumes actually does and is.
my vcenter and my vdi machine doesnt see same datastore . so is it problem for app volumes

Yes it is a problem. Your Appvolumes manager must talk to the VCenter server to reconfiigure the VDI machines. So your VDI machines must be created in the VCenter your Appvolumes servers is talking to.
Otherwise it can't attach the appstacks and writable volumes.
