I can un-assign multiple app stacks to user in one shot but assigning multiple app stacks in one shot I am not sure, may be .bat script (or) develop one API

Depends on what you are trying to achieve. Do you want to assign the appstack directly to the user? I would only suggest this for testing purposes, but yes you can.
In the manager go to Directory, then go to the tab Users. Search for the user you want to assign the appstack for. Keep in mind if you have a large user database this searching could take up to a minute.
Then select the user (you will see detailed info about the user, it's CN, last logged on info and stuff like that), then in the top right corner click Assign Appstack and you will get a new screen in which you can select the appstacks you want.

I am trying to do here, if I do "one click" the respected user have to get all app stacks [for example app stacks related to tester in my environment] may be I can achieve it from script?

Okay yes I'm not quite sure if that is an option to be honest. My guess is you need to run a script against the API, it might be an option.
Haven't tried to however.
You could explore the Groups tab in Directory...
You can assign multiple appstacks to the one group, then assign that one AD group to your user?
I like this strategy, unfortunately my environment is different [even user role as tester, they will be getting different app stacks based on tester work/usage]
