Since I do not have a large VMware infrastructure i have come to the conclusion that it is ok to have vCenter and SQL Express on the same server. What happens if this server dies? Will my VM's still function correctly? Should I be using a solution like DoubleTake to replicate this server to another server or are my tape backups suffice? If the server dies, can i just rebuild another server and install vCenter and SQL Express and start fresh? Thank you.

Your VMs will still work, but VC will be dead till you rebuild you SQL DB... During this time you can directly connect to your ESX if you need to power on/off your VM. If you are choosing SQL Express for your environment I would not bother with DoubleTake... Just have valid backup of you SQL DB and restore in case of disaster (yes, you might lose some performance data)
