Current config:
- 3 ESX 3.01 hosts
- Virtual center 2.5 u3
I'm going to upgrade all ESX 3.01 host to ESX 3.5 U3 using the Full installation CD image.
About 10 lun assigned to these ESX host.
Before perform the upgrade, should I disconnect all fiber connection first from ESX host?

If you're going to do a clean install. Yes, disconnect all HBA's on the ESX before performing the upgrade (it's recommended).
You should take a look at vCenter and update manager as well, nice tools.

I'll do a inplace upgrade from ESX 3.01 to ESX 3.05.

In case if you want to upgrade in place, we dont need to disconnect the HBA cards. Just that make sure while doing an upgrade you select the desired destination where system files are been installed earlier {mostly local hard drive}.

I would not do an upgrade but a reinstall. In this case definitely disconnect fibre unless you know how to do it via software but that is getting trickier to do.

I did an upgrade on my 5 hosts 3 weeks ago from ESX 3.01 to ESX 3.5 U3, and they have been running just fine w/o any issue.
I would definitely disconnect them. Although there might be no risk, it is better being safe than sorry.
